The identification of new biomarkers is an increasingly essential element of predictive, preventive and personalized medicine. To meet this need, The Biomarkers Consortium projects serve to develop promising biomarkers in order to help accelerate the delivery of successful new technologies, medicines and therapies for prevention, early detection, diagnosis and treatment of disease.

Working together, the members of the Biomarkers Consortium are building uniquely powerful collaborations that are increasing the development of biomarker-based technologies, medicines and therapies for the prevention, early detection, diagnosis and treatment of disease.

Biomarkers Consortium - Evaluation of the Utility of Adiponectin as a Biomarker for Predicting Glycemic Efficacy

The primary objective of this project was to determine whether a 30kDa adipocyte-secreted protein, adiponectin, has utility as predictive serum biomarker of glycemic control in normal non-diabetic subjects and patients with type 2 diabetes, following treatment with a novel and promising new class of compounds, PPARγ agonists. Results confirmed previous relationships between adiponectin levels and metabolic parameters, and support the robust and predictive utility of adiponectin across the spectrum of glucose tolerance.

Biomarkers Consortium - Placebo Data Analysis Project in Alzheimer’s Disease/Mild Cognitive Impairment Clinical Trials

The Biomarkers Consortium’s Placebo Data Analysis Project in Alzheimer’s Disease/Mild Cognitive Impairment Clinical Trials combined placebo data from large clinical trials provided by multiple pharmaceutical companies to create datasets of 3,000 to 5,000 subjects for Alzheimer’s disease (AD) and mild cognitive impairment (MCI) groups. The goal of the project was to develop better measures of disease progression in terms of outcome measures that have both low variability and are sensitive to change, for use in future clinical trials.

Biomarkers Consortium - Carotid MRI Development and Validation via an AIMHIGH Sub-Study

The goal of this project was to conduct a 75-patient study at a total of 15 centers to determine the reproducibility of the non-invasive technique of carotid magnetic resonance imaging (CMRI). Results established a standardized carotid MRI protocol and determined, for the first time, that kinetic parameters of carotid atherosclerotic plaque are reproducible and can be used for multi-center studies.

Biomarkers Consortium - I-SPY TRIAL-2 (Investigation of Serial Studies to Predict Your Therapeutic Response with Imaging and MoLecular Analysis): An Adaptive Breast Cancer Trial Design in the Setting of Neoadjuvant Chemotherapy

Phase II trial of novel investigative breast cancer agents in the neoadjuvant setting that uses biomarkers and adaptive design to accelerate the clinical trial process.

Biomarkers Consortium - Osteoarthritis Project

The Biomarkers Consortium - Osteoarthritis Project is a $3.4 million study aimed at determining which biomarkers have greater prognostic ability to measure early progression of structural and symptomatic changes in the joint over time and which are likely to predict treatment response better than the radiographic measurement of narrowing of joint space in knee OA patients. These new biomarkers are candidates for follow-on studies for evaluation and use in regulatory decision-making.

Biomarkers Consortium - Comparison of Two PET Radioligands to Quantify the Peripheral Benzodiazepine Receptor

The Biomarkers Consortium’s PET Radioligand Project, completed in December 2012, developed improved, more sensitive PET radioligands with higher binding to the peripheral benzodiazepine receptor. Findings from this study suggest that the [11C]PBR38 ligand, in particular, may be useful in detecting progression from mild cognitive impairment or treatment response in Alzheimer’s Disease.

Biomarkers Consortium - Establish Guidelines for Initial Diagnostic Criteria for “Sarcopenia with Clinically Important Weakness” and Associated Evidence for Treatment Benefit

The Sarcopenia 1 project launched in 2010 and aimed to establish the first evidence-based definition of sarcopenia (muscle weakness), which is still not recognized as a medical condition.

Biomarkers Consortium - Use of Targeted Multiplex Proteomic Strategies to Identify Plasma-Based Biomarkers in Alzheimer’s Disease

The Biomarkers Consortium’s Targeted Plasma-Based Biomarkers in Alzheimer’s Disease (AD), completed in July 2012, was the first part of a multi-phased effort utilizing samples from the Alzheimer’s Disease Neuroimaging Initiative (ADNI) to validate multiplex panels in both plasma and cerebrospinal fluid (CSF), to diagnose patients with AD and to monitor disease progression.

Biomarkers Consortium - Workshop: Developing an Evidentiary Criteria Framework for Safety Biomarkers Qualification

This workshop aimed at creating alignment among scientific stakeholders including the FDA, the NIH, the biopharmaceutical industry, academic researchers and patient groups regarding a proposed framework for determining the levels of evidence required to qualify biomarkers for use in drug development.
